After graduating from Syracuse University, Posnick began working at British Vogue before returning to New York where she became first a fashion associate, and eventually Beauty Editor, of American Vogue. In 1987, after having worked for Calvin Klein and Glamour magazine, Posnick returned to Vogue to work with Anna Wintour as Executive Fashion Editor, a position she kept until November 2018. She is now a Contributing Editor.

In 2009, Posnick edited Extreme Beauty in Vogue, a book showcasing some of the most ground-breaking images published in Vogue over the previous 75 years. The book was published in conjunction with an exhibition of the same title held in Milan. In 2016, Stoppers: Photographs from My Life at Vogue was published by Abrams. The book highlights the most exceptional editorials Posnick created throughout her 30-year career in addition to some behind-the-scenes stories from her time at Vogue.
